Creeper Woods Rune Room is located in a tombstone-like hole to the south-west in the caravan area of Creeper Woods. The button that opens the entrance door of Creeper Woods Rune Room will only appear after pressing the button at the back of Great Hall in Camp. Like other rune rooms, Creeper Woods Rune Room consists of a thin walkway suspended over an endless abyss of purple fog. Upon activating the button at the end of the walkway, several pillars will rise up in the background and the O rune will be able to be collected.

In the game files, Creeper Woods Rune Room uses the following objectgroup IDs in the following order: rr_entrance001, rr_tile001, rr_runeroom001, rralt_entrance001, rr_rockprop01, rr_rockprop02, rr_rockprop03, rr_rockprop04, rr_rockprop08, rr_rockprop07, rr_rockprop06, & rr_rockprop05
Strangely, the entrance door to this rune room will half-open simply by standing over it. It can be entered in this half-open state, and pressing the button will open it fully.
The entrance door of Creeper Woods Rune Room is one of only two instances of doors that are placed in the floor, with the other being the entrance to Hydrothermal Vent.
